Automation of sample preparation procedures in an Anti-Doping Laboratory is of great interest in terms of time saving, security inside the laboratory and eventually the quality of the results obtained. In order to automate the sample prep process, Zinsser has developed an automated pipetting system for liquid-liquid extraction, with further capabilities including heating, shaking, drying and crimping. It has been used for the automation of the method implemented for the determination of anabolic agents, narcotics, anti-estrogenic substances, cannabinoids and stimulants in urine by GC-MS and GC-MS/MS analysis, whose sample preparation procedure includes enzymatic hydrolysis, extraction and derivatization steps.

The system can completely automate all of the required sample steps. Here is a typical sample preparation method:

  • Adjust pH
  • Add phosphate buffer + internal standard + enzyme
  • Vortex
  • Incubate at 55 C
  • Add carbonate buffer + TBME for Liquid-Liquid Extraction
  • Shake
  • Phase separation
  • Evaporate to dryness
  • Add derivitization solution
  • Incubate at 65 C
  • (Ready for GC-MS or GC-MS/MS)

The system can prepare 96 samples for analysis overnight. The system has been tested and validated for 70 substances of interest by a major anti-doping laboratory, and is suitable for routine analyses of urine samples.
