Using the Sepmatix parallel HPLC system, eight samples can be processed
simultaneously to increase throughput. Samples are separated on an arrangement
of eight columns using a single pump. With the optional column oven each column can be independently temperature-controlled or even chilled. The flow of this pump is controlled and spread on the individual columns by the Sepmatix FlowControl, a patented active splitter module for measurement and regulation of the actual flow. The integrated Sepmatix 8x Autosampler is able to inject eight samples in parallel. Detection of all eight lines is done by one multiplexed PDA-Detector.

Even though HPLC technology is well established and widely
used today in many fields of pharmaceutical and biotechnology
research, there are physical limitations which affect the level of
throughput. Using large numbers of automated HPLC systems in parallel is not
an effective solution, because they are so costly, take up so much
space and require so much time and effort to operate. they also
require considerable maintenance. For example, if eight columns are used to separate the samples, then maintenance will not only
be required for these columns, but it will also be required for a
series of HPLC pumps, autosamplers and detectors.
HPLC systems that use higher pressure (over 400 bar) to deliver
quicker results have their own limitations. This is because they
need very pressure-resistant column materials in order to provide
good separation results – but many of the materials used for HPLC
technology are not yet available in a pressure-resistant form, or are
simply not suitable. as a result, many of these high-pressure HPLC
systems are used like conventional HPLC systems and therefore
do not contribute to increasing throughput.
The 8x parallel Sepmatix HPLC system from Sepiatec only uses
one external HPLC pump. The flow is separated by the patented Sepmatix FlowControl into eight channels, and the flow in each
channel is precisely measured and regulated throughout the entire
run. Together with an autosampler and an 8x parallel working detector,
the result is an unbelievably compact HPLC system which
will instantly enhance the throughput of every research and development
laboratory eight times over.
Because the Sepmatix works with standard columns and methods, it is not necessary to spend time adapting methods to the new
system or to purchase any new costly HPLC columns.

The Sepmatix 8x FlowControl module guarantees constant flow
rates in up to eight channels, while using just one HPLC pump. The FlowControl is a key part of the Sepmatix system,
but can be used as a stand-alone unit to adjust the flow of existing
HPLC or LC-MS systems if required. Sepiatec also offers a 4x FlowControl four channel unit. Both the
8x and the 4x FlowControl systems are available as an analytical
version (with fl ow rates from 0.25 to 1.5 ml/min per channel), a
semiprep version (1.5 to 6.0 ml/min per channel) and a prep
version (6.0 to 20.0 ml/min per channel).
Sepmatix Semi-preparative
With the preparative Sepmatix, an 8-fold fully automated separation and fractionation into either 96 standard well plates or deep well plates is possible. Fractionation is absolutely precise by use of the integrated peak detection algorithm.
Fractions from the eight channels are collected by a compact Fraction Collector, where every channel can be operated in a completely independent manner. The sophisticated design allows compact and precise loading of up to 12 microplates for each single channel. This results in up to 9216 collected fractions. Clear allocation of the corresponding wells makes it easy to harvest the individual fractions for further testing procedures.
Typical flow rates of the semi- preparative Sepmatix system are between 1,5 and 6,0 ml/min per column.
Sepmatix Analytical
The integrated Sepmatix 8x Autosampler is able to inject eight samples in parallel either from shallow or deepwell microplates.
The high level of automation permits the continuous processing of 10 deep well plates or 30 shallow well plates. Detection is based on a multiplexed PDA-Detector where the light of a deuterium UV light source is split into individual beams by separating the fiber bundle into eight individual channels.
